PURPOSE:To obtain a cytoplasmic type tyrosine kinase, useful for evaluating or controlling the degree of differentiation of blood cells, a carcinostatic agent, genetic therapy, etc., and capable of controlling the differgntiation of the blood cells and a DNA capable of coding the tyrosine kinase. CONSTITUTION:This new tyrosine kinase is a polypeptide having an amino acid sequence expressed by the formula and a DNA is the one coding the polypeptide. This DNA is obtained by cloning a tyrosine kinase related to the differentiation of a human megakaryoblast leukemic cell strain UT-7 according to a reverse transcription-polymerase chain reactional (RT-PCR) method and providing the DNA capable of coding the total length from a cDNA library of the UT-7 using a tyrosine kinase gene fragment increasing the extent of expression with the differentiation as a probe. Thereby, the base sequence of the DNA is determined to prepare an anti-sense DNA and an RNA and a transformed cell, etc., from the resultant gene. |
