PURPOSE:To provide the subject new microorganism belonging to Bacillus sp. KS-425 stipulated as Bikoken stipulation no. 9007, exhibiting alkali cellulase K-425 productivity, growing in a neutral medium and capable of producing an enzyme excellent in its activity. CONSTITUTION:A dispensing spoon (about 5g) of the soil in Ichigai cho, Haga gun, Tochigiken prefecture is suspended in a sterilized physiological salt solution and heat treated at 80 deg.C for 10min. The supernatant of this heat-treated solution is subsequently suitably diluted and applied to an agar-agar culture medium for separation. Culture is carried out at 30 deg.C for 3 day to form a colony and a microorganism forming a transparent zone due to dissolution of carboxymethyl cellulose(CMC) around the colony is then selected to collect a CMCase-producing microorganism. The obtained CMSase-producing microorganism is inoculated in a liquid culture medium and subjected to shaking culture at 30 deg.C for 3 day. The cultured solution is subsequently centrifuged and the resultant supernatant is subjected to detection of CMCase activity at pH 3 to 13 for screening of an alkali cellulase-producing microorganism. Thus, the objective Bacillus sp. KSM-425 (Bikoken stipulation no. 9007) exhibiting alkali cellulase K-425 productivity can be obtained. |
