PURPOSE: A diagnosis method of Pseudomonas syringae pv. actinidiae is provided for early and simple detection. CONSTITUTION: The method comprises the steps of: identifying P. syringae pv. actinidiae-specific DNA fragment (about 670bp); transforming and culturing E. coli HB101 with pGEM-T vector containing the DNA fragment (KCTC 8895P); determining the DNA sequence of the fragment (668bp); confirming P. syringae pv. actinidiae strains by Southern blot analysis with a specific DNA fragment as a probe; designing forward and reverse primers based on the DNA sequence; diagnosing bacterial pathogen by colony-PCR.
