PROBLEM TO BE SOLVED: To obtain a new DNA decomposing enzyme consisting of a protein having a specific amino acid sequence or a decomposing enzyme specific to a DNA sequence as a functional mutant thereof, having a high thermal stability and useful as a reagent for the gene technology, etc. SOLUTION: This new DNA decomposing enzyme consisting of a protein having an amino acid sequence expressed by the formula or a decomposing enzyme specific to a DNA sequence as a functional mutant thereof, binding with a DNA having a cross-shaped structure, having a high thermal stability and useful as a reagent for a gene technology, etc., is obtained by separating a genomic DNA from Pyrococcus furiosus, treating the DNA with a restriction enzyme, incorporating the treated DNA into a cosmid vector for forming a cosmid library, then heat-treating to form a heat resistant protein library, screening the library with a gel shift assay, incorporating the obtained gene into an expression vector, transforming host cells with the expression vector and culturing the transformed cells. |
