A recombinant plasmid capable of replication in Escherichia coli and Bacillus sp. is provided, which plasmid is isolated from Bacillus sp. having antifungal activity, and is able to be replicated in E. coli and Bacillus sp. without introduction of a gene isolated from E. coli, so that the plasmid can be useful for cloning of various foreign genes in E. coli and Bacillus sp. A plasmid pWBL5 is isolated from Bacillus lentimorbus WJ5(KFCC-11316) having antifungal activity and has the nucleotide sequence set forth in SEQ ID NO:1. The recombinant plasmid capable of replication in Escherichia coli and Bacillus sp. comprises a portion or the total of the plasmid pWBL5, and a selection marker, wherein the selection marker is selected from an ampicillin resistance gene, a kanamycin resistance gene, a streptomycin resistance gene, a tetracycline resistance gene, an erythromycin resistance gene, and a chloramphenicol acetyltransferase gene. The method for preparing the recombinant plasmid pWBL5EBS capable of replication in Escherichia coli and Bacillus sp. comprises digesting the plasmid pWBL5 and a vector pBluscript SK(+) with a restriction enzyme PstI, and ligating the digested plasmid pWBL5 with the digested vector pBluscript SK(+), wherein the recombinant plasmid pWBL5EBS has a cleavage map of figure 5.
