A cell culture media for plasmid production is provided for increased production of a recombinant GDF-5 (rhGDF-5) protein. Also provided herein is a method of making a cell culture media for plasmid production and a method for growing a plasmid vector. The methods of enhancing production and protein expression of rhGDF-5 protein as disclosed are cost-effective, time-saving and are of manufacturing quality.

A cell culture media for growing a cell that expresses recombinant human growth and differentiation factor-5 (rhGDF-5) protein, the cell culture media supporting growth of a cell that comprises an expression vector having a polynucleotide encoding an rhGDF-5 protein under the control of a T5 or Trc promoter, and wherein the pH of the cell culture media ranges from about 6.5 to about 7.1, and the cell culture media does not contain glucose, and the rhGDf-5 protein comprising SEQ ID NO:4.
