Disclosed are a biosensor and a manufacturing method thereof. The biosensor comprises: a substrate; a metal layer formed on the substrate; a dielectric layer formed on the metal layer, having a low refractive index having a reference value or less; and a nanostructure layer which has a plurality of nanostructures formed on the dielectric layer. As such, the biosensor maintains high-sensitive properties by forming the dielectric layer between the metal layer and the nanostructure layer.
