PURPOSE:To accurately measure an amount of bacterium belonging to the genus Methanosarcina, acetic acid assimilating methane bacterium by determining an amount of alpha-hydroxyarchalol in a test specimen. CONSTITUTION:An amount of alpha-hydroxyarchoalol in a test specimen is measured. Preferably, an alpha-hydroxyarchalol-containing ether bond type lipid is extracted from the test specimen, a polar group in the liquid is eliminated give an alpha-hydroxyachalol-containing neutral lipid, to which a labeled substituent group is introduced, made into a derivative and alpha-hydroxyarchalol made into the derivative is detected. Measurement of the alpha-hydroxyarxhalol made into the derivative, for example, is carried out by treating making alpha-hydroxyarchalol with anthroylnitrile into the derivative and measuring its fluorescence. |
