PROBLEM TO BE SOLVED: To provide a method for simply determining or detecting DNA, and the like. SOLUTION: The method for determining or detecting the content of a methylated DNA in a target region in DNA contained in a specimen, includes (1) a first process for obtaining base sequence-containing DNA in the target region from DNA contained in a specimen, (2) a second process for treating the DNA obtained in the first process with a methylating enzyme to obtain the methylated DNA, (3) a third process for obtaining a single strand methylated DNA from the methylated DNA obtained in the second process, (4) a fourth process for binding the single strand methylated DNA selected in the third process to a methylated DNA antibody and a specific oligonucleotide to form a complex of the target methylated DNA region-containing single strand methylated DNA, the methylated DNA antibody and the specific oligonucleotide, and then obtaining the complex, and the like; and others are also provided.
