The invention relates to the regulation of the body weight of animals, including mammals and humans, in particular to materials indicated as weight modulators, and to the potentials of applying suchmodulators in diagnostics and therapy. The invention relates to the discovery and characterization of nucleotide sequences and of proteins about which it is presumed that are expressed by such nucleotides or their variants manifesting their ability to participate in the regulation of mammal body weights. The nucleotide sequences are the genes corresponding to the mouse or human OB gene which is considered to play a key role in the regulation of the body weight and for obesity. According to preliminary data the polypeptide product of the said gene functions are a hormone. The invention also relates to the application of nucleic acid molecules as probes or primers for amplification by polymerase chain reaction (PCR), i.e. synthetic or natural oligonucleotides, to cloning vector containing the nucleic acids, according to this invention, to expression vector in bacteria or in insect or mammal cells and to vector containing the nucleic acids functionally related to a sequence controlling the expression. The invention also relates to a bacterial cell or a mammal cell transfected or transformed by a suitable expression vector, to the application, respectively, to the said structures to antibodies against the OB polypeptides and to a method for the modulation of the body weight of mammals. |
