The present invention relates to a sample pre-treatment method using calcium oxide, wherein the method comprises the steps of: treating and mixing a sample with calcium oxide; leaving alone the sample mixed with calcium oxide; extracting genes from the sample left alone from the previous step; and amplifying the extracted genes and analyzing the genetic diversity thereof. According to the present invention, it was found that when performing the genetic detection of biospecies in a soil sample, a sample untreated with calcium oxide has a low detection sensitivity to minority microbial populations due to the presence of dominant species, whereas a sample treated with calcium oxide has a decreased population of dominant species such that a variety of biospecies that are undetected in the untreated sample are detected. Accordingly, the sample pre-treatment method using calcium oxide of the present invention can enhance the detection sensitivity to less dominant species, and thus can be used in developing a pretreatment reagent product or applied to an existing genetic detection kit. |
