The present invention relates to a nucleic acid molecule for recombinant expression and secretion of a peptide or protein of interest comprising a hemolysin A and/or hemolysin C-derived nucleotide sequence, fragments thereof, homologs thereof, or the complements thereof, and a nucleotide sequence encoding the peptide or protein of interest.

1. An isolated nucleic acid molecule comprising a first nucleic acid sequence operably linked to a second nucleic acid sequence, wherein:
(a) the first nucleic acid sequence comprises a nucleotide sequence having at least 70% nucleotide sequence identity to any one of the nucleotide sequences as set forth in SEQ IDs NOs: 1 and 3-27 in which SEQ ID NO: 2 is present, or the complements thereof, and (b) the second nucleic acid sequence is located 3′ to the first nucleic acid sequence and comprises a polynucleotide encoding at least one peptide or protein of interest heterologous to a hemolysin Type I secretion system.
