ADSORPTION OF NUCLEIC ACID ON SOLID PHASE

PROBLEM TO BE SOLVED: To provide an alternative method in which a biological sample containing a nucleic acid is prepared and the nucleic acid is adsorbed on a solid phase, and which method overcomes alcohol requirement in the absorption step. SOLUTION: The method for adsorbing on the solid phase the nucleic acid derived from the biological sample comprises (a) a step for providing an aqueous solubilization buffer solution containing a chaotropic agent; (b) a step for mixing the aqueous solubilization buffer solution with a biological sample so that the concentration of the chaotropic agent is 1-4 mol in the mixture, and incubating the mixture; (c) a step for dissolving an additional amount of chaotropic agent or adding an aqueous solution containing the additional chaotropic agent so as to increase the chaotropic agent in the mixture by 0.5 mol or more; and (d) a step for contacting the mixture of the (c) step with the solid phase to adsorb the nucleic acid derived from the mixture on the solid phase.
