Provided is a method for determining the gene state of a fetus, said method comprising: a target region selection step for selecting a target region for determining the gene state from among regions on human genome; a step for isolating a single cell from a maternal blood sample; a step for extracting genomic DNA from the single cell; a step for PCR amplifying the target region using the genomic DNA extracted from the single cell as a template and employing a primer set that is designed so as to PCR amplify the target region; and a DNA sequencing step for decoding the DNA base sequence of the PCR amplification product of the target region. In this method, the aforesaid primer set designed so as to PCR amplify the target region has been designed by a method for designing a primer set to be subjected to a polymerase chain reaction, said method comprising a first stage screening step based on local alignment scores and a second stage screening step based on global alignment scores. |
