PURPOSE: To enable detecting a cobalamin or folic acid deficiency in the body fluid of a warm-blooded animal by measuring the level of the total amount of homocysteines in the body fluid. CONSTITUTION: Human blood serum, for example, is mixed with an internal reference standard labeled by an appropriate marker containing a known amount of homocysteine species to be measured, and all of labeled or unlabeled homocysteines are reduced to a free state and homogenized by adding a reducing agent such as 2-melcaptoethanol. Next, metal ions are removed by addition of a chelating agent as necessary, the reaction mixture obtained is strained and heated at about 25 to 150 deg.C for about 1 to 60 minutes, and after randomization of the labeled and unlabeled homocysteines is ensured, their amounts are measured and their ratio is calculated and compared with a standard known amount to determine the total amount of endogenous homocysteines present in the human blood serum. The increased level of the total amount of homocysteines enables precise diagnosis on cobalamin and folic acid deficiencies. |
