Nuclear receptor and mutant thereof and the use of the same in the reprogramming of cells

According to the invention there is provided methods for inducing pluripotent stem cells in vitro, comprising introducing a gene or polypeptide of a nuclear receptor and one or more gene or polypeptide selected from the group consisting of Sox, Krüppel-like factor or the myc family, to cells in vitro. The present invention also provides vectors and compositions for producing the same and methods for using the induced pluripotent stem cell for treating a patient in need of a pluripotent stem cell treatment.

1. A method for inducing pluripotent stem cells comprising the steps of:
(a) culturing cells in vitro; (b) introducing, into cultured cells of step (a), exogenous reprogramming factor genes, wherein the exogenous reprogramming factor genes consist of
(i) Nr5a2 or a sumoylated mutant thereof, Sox2 and Klf4;(ii) Nr5a2 or a sumoylated mutant thereof, Sox2, Klf4 and c-Myc; or(iii) Nr5a2 or a sumoylated mutant thereof, Nr5a1 or a sumoylated mutant thereof, Sox2, Klf4 and c-Myc, thereby inducing pluripotent stem cells in vitro.
