PROBLEM TO BE SOLVED: To provide a caspase inhibitor useful for the treatment of many disorders such as cancer, acute inflammation and autoimmune injury, ischemic disorder, and specific neural degeneration disorder. SOLUTION: This caspase inhibitor is a compound represented by formula I, wherein R<SP>1</SP>is CO<SB>2</SB>H, CH<SB>2</SB>CO<SB>2</SB>H, or their esters, amides, or isosteres; R<SP>2</SP>and R<SP>3</SP>are each independently selected from hydrogen or a 1-6C aliphatic group, which is substituted if desired; and R<SP>4</SP>and R<SP>5</SP>are each independently selected from hydrogen or a 1-6C aliphatic group, which is substituted if desired, or R<SP>4</SP>and R<SP>5</SP>each forms a bicyclic ring, which is substituted if desired, together with the ring with which they are bonded.
