PROBLEM TO BE SOLVED: To provide a method for optically detecting glucose and the like in the body fluid, and to provide a detection reagent suitable for the purpose. SOLUTION: The method for optically detecting an analyte in a sample comprises (a) a step wherein the sample is brought into contact with a detection reagent comprising an nicotinamide-dependent oxidation reduction enzyme, a reducing nicotinamide coenzyme, a mediator comprising quinone and a reducing optical indicator or indicator system so that the analyte is oxidized with the enzyme and the enzyme is reduced and the electron of the reduced nicotinamide coenzyme is transferred to the optical indicator (system) with the mediator, and (b) a step wherein existence and/or amount of the analyte in the sample is measured by optically detecting the indicator (system).
