Provided are method for inducing immune tolerance to antigens including therapeutic antigens. The method comprises administering to an individual OPLS or other lipidic compositions and the antigen, either in a single formulation or as separate formulations where the separate formulations are administered concomitantly or sequentially.

1. A method for inducing immune tolerance toward an antigen comprising:
a) providing a first composition comprising O-phospho-L-serine (OPLS), and a second composition comprising the antigen, wherein the first composition is substantially free of the antigen and the second composition is substantially free of OPLS; b) administering the first and the second compositions separately through a cutaneous route to an individual; wherein the two compositions are administered at sites that are in close proximity of the each other, wherein administration of OPLS and the antigen results in induction of immune tolerance in the individual toward the antigen.
