EXPRESSION SYSTEM FOR STEM LOOP TYPE RNA MOLECULE HAVING RNAi EFFECT

PROBLEM TO BE SOLVED: To provide a new expression system for an RNA molecule in which expression of a target gene can be suppressed and to provide a method for preparing a knockdown cell using the system.

SOLUTION: It is found that a stem loop type RNA molecule can efficiently be translocated into cytoplasm and RNi (RNA interference) effects can be exhibited by producing the RNA molecule from a DNA encoding the stem loop type RNA molecule utilizing a tRNA promoter. The RNAi effects can efficiently be exhibited even by transferring a cytoplasm translocation signal sequence into the DNA encoding the stem loop type RNA molecule. A cell in which a desired gene is knocked out can simply be prepared by using the expression system for the stem loop type RNA molecule.
