PROBLEM TO BE SOLVED: To efficiently obtain the subject library by forming RNA-DNA conjugate by reverse transcription using mRNA as a template and bonding a tagged molecule to diol structure on 5' side of mRNA and separating DNA. SOLUTION: This method comprises preparing library of cDNA corresponding to complete length of mRNA. RNA-DNA conjugate is formed by reverse transcription from a primer by using mRNA as a template and a diol structure which exists on the 5' Cap (<7> M<e> GPPPN) site of mRNA forming the RNA-DNA conjugate is subjected to oxidative ring-opening reaction with sodium periodate to afford dialdehyde and a tag molecular having hydrazine end is reacted with the dialdehyde to chemically bond the dialdehyde to the tag molecular and RNA-DNA conjugate having DNA corresponding to complete length of mRNA among RNA-DNA conjugates to which tagged molecule is bound is separated by utilizing function of tag molecule to provide the objective cDNA library. |
