DEVELOPMENT OF MOLECULAR MARKER ASSOCIATED WITH BACKFAT THICKNESS AND MARBLING IN HANWOO

Molecular markers associated with backfat thickness and marbling in Hanwoo(Korean cattle) are provided to improve rapidness, convenience and accuracy of the screening and breeding for a Hanwoo having high backfat thickness and marbling in Hanwoo group by using a promoter SNP(single nucleotide polymorphism) marker of an obesity gene . Molecular markers associated with backfat thickness and marbling in Hanwoo(Korean cattle) are detected by isolating genomic DNA from a biological sample of Hanwoo including muscle tissue, blood and hair root; amplifying a specific site of a leptin gene promoter in the isolated DNA through PCR(polymerase chain reaction) by using a primer of SEQ ID NO:1; sequencing the amplified PCR product through direct sequencing to detect two SNPs; analyzing SNP genotypes of the leptin gene promoter by using PCR-SSCP(single strand confirmation polymorphism) to detect five kinds of SSCP genotypes including AA, BB, CC, AC and BC, wherein AC genotype of Hanwoo is determined as having high backfat thickness and marbling.
