PURPOSE:To obtain a uniformly labeled <13>C-labeled amino acid useful for the investigation of metabolism in vivo can an industrial scale at a low cost by automatically culturing a methane assimilating microorganism in a medium containing <13>C-labeled methane in a perfectly closed system, extracting a protein from the microbial cell and hydrolyzing the protein. CONSTITUTION:A uniformly labeled <13>C-labeled amino acid useful for the analysis of steric structure of protein or the investigation of metabolism of a protein in vivo is produced on an industrial scale at a low cost without leaking <13>C- labeled carbon dioxide gas from the system by filling a nutrient medium in a reactor 1 provided with a stirrer 2, a pH sensor 3 and a dissolved oxygen sensor 4, adding methane-assimilating microorganisms to the medium, circulating gaseous components such as <13>C-labeled methane, oxygen, etc., through circulation lines 7, 12, automatically culturing the microorganism in a perfectly closed system replenished with <13>C-labeled methane and oxygen by a gas pressure-swing absorber 13, recovering the cultured microbial cells, extracting and recovering the protein from the obtained cell and hydrolyzing the recovered protein. |
