RECOMBINANT TYPE II COLLAGENASE FROM CLOSTRIDIUM HISTOLYTICUM AND USE THEREOF FOR ISOLATION OF CELL AND CELL GROUP

New process to dissolve cell tissue and release cells by: (a) incubation with collagenase II from Clostridium histolyticum characterised in that it: (i) has substantial activity after 12 rounds of freezing; (ii) has the partial 162 amino acid sequence encoded by the 487 bp nucleotide sequence (I) (given in the specification); and (III) is the expression product of prokaryotic or eukaryotic exogenous DNA; and (b) separation of the isolated cells. Also claimed are: (1) prodn. of a polypeptide with collagenase class II characteristics as above; (2) (I) as above; (3) a plasmid or viral vector contg. (I); and(4) prokaryotic or eukaryotic cells stably transformed with the vector as in (3).
