<P>PROBLEM TO BE SOLVED: To provide a method and a composition for differentiating stromal cells from adipose tissue into cells having osteoblastic properties, and a method for improving a subject's bone structure. <P>SOLUTION: The method comprises a process for culturing the stromal cells from the adipose tissue in β-glycerophosphate and ascorbic acid and/or ascorbate-2-phosphate for a time sufficient to allow differentiation of the cells into osteoblasts. The method and the composition like this are useful for producing the osteoblasts for autologous transplantation into a bone at a surgical site or injury. This composition comprises adipose stromal cells, a medium able to support the growth of fibroblasts and sufficient amounts of β-glycerophosphate and ascorbic acid and/or ascorbate-2-phosphate or both of them to induce the differentiation of the stromal cells into osteoblasts. Further, a method for identifying compounds affecting the differentiation of osteoblasts is provided.
