MANUFACTURING METHOD OF ANGIOTENSIN CONVERTING ENZYME INHIBITOR FROM OLD PUMPKIN

PURPOSE: A manufacturing method of an angiotensin converting enzyme(AEC) inhibitor from an old pumpkin is provided, to develop a new high functional food material for the treatment of high blood pressure by using hydrothermal extraction, vacuum concentration, ultrafiltration with a molecular weight limit of 1000 Dalton, gel permeation chromatography(GPC), and reversed-phase high pressure liquid chromatography(reversed-phase HPLC). CONSTITUTION: The pumpkin is washed and cut for gyre extraction at 95°C for 2 hours. The extract is filtered through a filtering paper and concentrated in vacuum by using a rotary vacuum evaporator at 65°C. The dried concentrate is dissolved in 0.01M of phosphate buffer solution (pH7.0) after lyophilization and is centrifuged at 12,000 rpm and 4°C for 30 min. The upper part of the solution after centrifuging is ultrafiltered and the filtered solution is loaded on a gel permeation chromatography(GPC). The column of the GPC uses resin of Sephadex G-15 and the phosphate buffer solution at pH 7.0. The ACE inhibitor is separated from the lung of a rabbit and uses Hippuryl-L-histidyl-L-leucine(HHL) as a substrate.
