发明名称 PROMPT DISCRIMINATION OF FUNGI, BACTERIA AND YEAST
摘要 PROBLEM TO BE SOLVED: To provide a discrimination process that can promptly and exactly distinct bacteria comprising prokaryotic cells and can simply and exactly distinguish fungi and yeast comprising eukaryotic cells by rationalizing the selective permeation of a fluorescent dye into these cells. SOLUTION: A fluorescent dye 2B as fluoresceine or its derivative and another fluorescent dye 2C as propidium iodide are mixed in a concentration of 3-15μmol/ml, respectively and the solution 2 is heated at 25-36 deg.C. Then, bacteria are dyed with this solution for 3-9 minutes and the fungi and yeasts are dyed for 15 to 30 minutes. Then, individual microorganism-dyeing solution 3 are irradiated with the exciting ray of 488 nm wavelength whereby the kinds of microorganisms, viable cells, dead cells and number of the cells are visually and promptly discriminated from the fluorescent emission from these viable cells and dead cells of these microorganisms.
