PROBLEM TO BE SOLVED: To enable acquisition of clear images of a specimen over a wide range even when a large focal depth cannot be secured.SOLUTION: A microscope 1 includes a detection optical system 7 that detects fluorescent light from a specimen S and acquires fluorescence images; a sheet illumination optical system 3 that directs planar excitation light in a direction crossing an optical axis of the detection optical system 7 toward the specimen S; and an image processing unit 20 that processes images acquired by the detection optical system 7. The sheet illumination optical system 3 moves a focal position of the excitation light along an incident direction thereof, and the image processing unit 20 merges a plurality of fluorescence images acquired by the detection optical system 7 to generate a merged image every time the excitation light is directed at a different focal position by the sheet illumination optical system 3.SELECTED DRAWING: Figure 1 |
