METHOD FOR DETECTING COPPER ION

PROBLEM TO BE SOLVED: To provide a new method for detecting slight amounts of copper ion. SOLUTION: The method for detecting copper ion in a sample comprises the step(1) of preparing a variant V1-ATPase, a conjugate molecule having three polypeptides in the Sequence No.3, three polypeptides in the Sequence No.4 and one polypeptide in the Sequence No.5 and having (a) the amino acid substitution of all of the Cys residues in the Sequence No.3 and the Sequence No.4 to Ser residues and (b) the amino acid substitution of the 232nd Ser residue in the Sequence No.3 to a Cys residue and/or the amino acid substitution of the 235th Thr residue to a Ser residue and the step(2) of contacting a sample solution that may contain copper with the variant V1-ATPase.
