The disclosure provides monoclonal bispecific antibodies targeting HER2 and HER3. The disclosure also provides monospecific tetravalent HER3 antigen binding antibodies. Still further provided by the disclosure are methods of treating a cancer in a subject, comprising administering to the subject a therapeutically effective amount of an antibody provided by the disclosure.

An isolated monoclonal, bispecific antibody comprising: (a) an anti-HER2 antibody comprising two heavy chains and two light chains and at least one anti-HERS single chain antibody variable fragment (Fv) comprising a heavy chain variable domain and a light chain variable domain linked to any one of the two heavy chains or two light chains of the anti-HER2 antibody, or (b) an anti-HER3 antibody comprising two heavy chains and two light chains and at least one anti-HER2 single chain antibody variable fragment (Fv) comprising a heavy chain variable domain and a light chain variable domain linked to any one of the two heavy chains or two light chains of the anti-HER3 antibody.
