Provided is a digital holographic microscope combining a fluorescence digital holographic microscope and a phase image detection digital holographic microscope and capable of simultaneously three-dimensionally measuring a fluorescence image and a phase image. The digital holographic microscope is provided with: a first holographic optical system which, using first laser light 5, acquires a phase three-dimensional image due to interference light generated by superimposing object light that passes through a sample stage 4 and reference light that does not pass therethrough on each other; a second holographic optical system which, using second laser light 6, acquires a fluorescence three-dimensional image due to fluorescence excitation light; an imaging means 3; and a bifocal Fresnel lens (Sub-wavelength Periodic structured Fresnel Lens (SPFL)) 2 having polarization dependency. The respective optical systems are disposed such that the object light and fluorescence signal light are coaxially superimposed, and the bifocal lens 2 is disposed on superimposed optical axes. The imaging means 3 simultaneously acquires the phase three-dimensional image and the fluorescence three-dimensional image as a hologram. The phase three-dimensional image and the fluorescence three-dimensional image are separated, and the object light and the fluorescence signal light are reconstructed from the interference intensity distributions of the respective three-dimensional images. |
