PROBLEM TO BE SOLVED: To obtain a DNA (fragment) which contains human bone-inducing factor-2 promotor domain having a specific base sequence, allows screening of a low molecular compound which controls expression of human bone-inducing factor-2, and is useful for preventing and treating, for example, bone diseases and excessive osteogenesis syndrome. SOLUTION: This DNA (fragment) contains human bone-inducing factor-2 promotor domain having a base sequence from base 1 to base 6728 shown by formula I. The DNA shown by formula I is obtained by (1) screening a P1 phage human genomic library by the PCR method using primers shown by formulas II anti III, (2) cleaving the isolated P1 phage with restriction enzymes SalI and SpeI, (3) identifying the objective fragment by Southern blotting, followed by (4) subcloning of the obtained objective fragment to pBluescript vector cleaved by the enzyme. It is preferable to carry out screening of bone- related substances using the whole DNA shown by formula I or a DNA prepared by linking a part of it to a reporter gene. |
