This disclosure provides a single reactor that accommodates an affinity selector to separate analytes of interest, and an enzyme reactor that digest the analyte to suitable peptides for mass spectrometry. The single reactor formats described herein accommodate workflows wherein separation precedes digestion as well as workflows wherein digestion precedes separation selection.

1. A single reactor wherein said single reactor performs both affinity purification and enzymatic digestion of protein samples, comprising:
an affinity selector, wherein said affinity selector retains and purifies at least one analyte of interest through physical properties of said analyte of interest; and an enzyme reactor, wherein said enzyme is kept relatively inactive during said affinity selection.
