1. A method of separating bacterial species and bacterial serotypes present in a sample comprising,
(a) providing a direct current insulator gradient dielectrophoresis (DC-iGDEP) device, the device comprising one or more members defining one or more separation chambers,
each separation chamber defining a microfluidic channel having a top, bottom and two sides, the channel having a channel inlet port at a proximal end and a channel outlet port at a distal end, and further comprisingan electrode disposed at the proximal end of the channel,an electrode disposed at the distal end of the channel,one or more sets of teeth, each set comprising two teeth disposed opposite from one another at the sides of and extending into the microfluidic channel and defining a gate in the channel, wherein the gates defined by the one or more sets of teeth are 1 to 500 microns across and establish local gradient regions that act as separate dielectrophoretic traps, each tooth having a base disposed at a side of the microfluidic channel and a tip disposed at a gate, (b) loading a loading volume of the sample into the channel inlet port, (c) applying an electrical field across the electrodes in the one or more separation chambers to separate the bacterial species or bacterial serotypes in the sample, and (d) identifying the bacterial species or bacterial serotypes present within the sample based on their behavior in the field present in the one or more channels. |
