#CMT# #/CMT# Detecting pathogenic Legionella microorganisms in water from a pipe system comprises filtering at least 20 liters of water through a filter connected to the pipe system, disconnecting the filter, releasing DNA from microorganisms collected on the filter with a protein-containing lysis buffer, and amplifying Legionella-specific fragments either by PCR and gel electrophoresis or by real time PCR. #CMT#USE : #/CMT# The method is useful for early detection of Legionella microorganisms in water samples (claimed), especially drinking water samples. #CMT#ADVANTAGE : #/CMT# Legionella microorganisms can be detected at levels below 100 microorganisms per 100 ml. #CMT#ENVIRONMENT : #/CMT# Preferred Method: The amount of water filtered is at least 50 liters. The microorganisms are collected on a deep filter, dispersed in a wash medium, concentrated and collected on a surface filter. The lysis buffer contains a detergent. The DNA is purified using a DNA isolation kit and prepared in small volumes suitable for PCR. |
