NOVEL THERMOPHILIC SUBTILISIN-LIKE SERINE PROTEASE DNA SEQUENCE AND PROTEIN DERIVED THEREFORM

PURPOSE: Provided is a DNA sequence of novel thermophilic subtilisin-like serine protease, which is isolated from Thermoanaerobacter younseii. Also, amino acid sequence translated therefrom is provided. CONSTITUTION: A DNA sequence of new thermophilic subtilisin-like serine protease is isolated from thermopile, Thermoanaerobacter yonseii(KFCC 11116) and represented by the figure (4b). Sequencing method of thermophilic protease DNA sequence and amino acid sequence thereof comprises the steps of: isolating genome DNA of Thermoanaerobacter younseii and manufacturing DNA thereof; synthesizing an oligonucleotide primer using the DNA library for screening thermophilic subtilisin-like serine protease; screening genomic DNA library; sequencing DNA sequence and analyzing it. PCR primer and probe which are used in DNA library screening are characterized by comprising the following: Pro I F : 5' AAY GGN CAY GGN CAN CAY 3' Pro I R : 5' YGG YGT YGC CAT NGA NGT NCC 3'.
