PROBLEM TO BE SOLVED: To provide methods of producing pluripotent stem cells from adult cells, in particular, methods of producing pluripotent stem cells from somatic cells without the use of a feeder-cell layer or an agent that increases efficiency of retroviral transfection.SOLUTION: Somatic cells plated on a tissue culture substrate are transduced with a retrovirus containing nucleic acid encoding at least one gene selected from the group consisting of SOX2, OCT4, LIN28 and NANOG, and cultured supplemented with an agent that inhibits Rho kinase activity and bFGF.SELECTED DRAWING: None |
