PROBLEM TO BE SOLVED: To obtain a DNA (fragment) which contains human bone-inducing factor-7 promotor domain having a specific base sequence, allows screening of a low molecular compound which controls expression of human bone-inducing factor-7, and is useful, for example, for preventing and treating bone diseases and renal disorder. SOLUTION: This DNA (fragment) contains human bone-inducing factor-7 promotor domain having a base sequence from base 1 to base 10877 shown by the formula. The DNA (fragment) is obtained by (1) digesting a human placenta genomic DNA with restriction enzyme HindIII, (2) isolating fragments by agarose gel electrophoresis, (3) cloning the isolated HindIII-digested DNA fragments to lambda phage vector λDASH-II treated with the enzyme, (4) packaging the obtained vector into the phage, (5) infecting Escherichia coli with the obtained phage, (6) preparing a genomic DNA library, (7) screening by the PCR method, followed by (8) subcloning to a plasmid vector. It is preferable to prepare a recombinant expression vector by incorporating, for example, the whole DNA shown by the formula into a reporter gene. |
