The present invention discloses the method of in vitro detecting aberrant dysplasia using methylation of p16CpG island nucleotides, The method comprises extracting target cell DNA from the tissue or liquid of body; detecting the methylation state of the sequences of p 16 CpG island nucleotides; determining the aberrant dysplasia in the tissue to be tested based on the sequences of methylated p16CpG nucleotides. The artificial sequences of nucleotides of p16CpG island being used were disclosed.
