PROBLEM TO BE SOLVED: To provide a detection method of a base substitution which is capable of efficiently screening base substitution technique of mRNA.SOLUTION: A method for detecting base substitution by substitution means of base to be evaluated includes: a process of executing the substitution means of base to be evaluated to a base substitution GFP-mRNA, in which a base substitution GFP gene, which a base in a base sequence of a GFP gene is substituted and GFP (green fluorescent protein) cannot be expressed, is transferred, re-substituting a substituted base to obtain GFP-mRNA; a process of translating the GFP-mRNA to obtain GFP (green fluorescent protein); and a process of detecting green fluorescent emission of the GFP.SELECTED DRAWING: Figure 2
