PROBLEM TO BE SOLVED: To provide a specimen reaction method to automatically perform the reaction of a specimen with a reagent stepwise (in multiple steps) using a small amount of the specimen and a small amount of the reagent to effectively reduce the cost of the specimen and the reagent and reduce the load on an experimenter when repeating the reaction. SOLUTION: The method for performing the reaction of a specimen with a reagent uses a substrate having a hydrophilic region and a hydrophobic region on the surface. Concretely, the specimen is mixed with the reagent in the hydrophilic region, the mixed specimen solution composed of the specimen and the reagent is covered with a sealing oil, the specimen solution is made to react under a prescribed temperature condition, the reaction product is separated from the hydrophilic region, the separated reaction product is mixed with the reagent in other hydrophilic region, the mixed reaction solution composed of the reaction product and the reagent is covered with a sealing oil, and the reaction solution is made to react under a prescribed temperature condition.
