PROBLEM TO BE SOLVED: To provide a method of shortening a process to prepare spheroids of established hepatocytes in screening a compound, and to obtain spheroids having drug metabolizing activity.SOLUTION: A screening method of a compound comprises the steps of: preparing spheroids 9 having equivalent diameter of 50 μm to 200 μm by culturing established hepatocytes in a culture vessel 10 for a period of 12 to 72 hours; incubating the spheroids 9 by adding to it one culture medium selected from the group consisting of first culture medium free of a test compound, second culture medium comprising a desired concentration of the test compound, and third culture medium comprising at least one inhibitor of a metabolic enzyme that may metabolize the test compound and a desired concentration of the test compound; measuring the viability of the spheroids 9 that were cultured by adding one of the first to the third culture medium; and determining whether or not the test compound has toxicity to hepatocytes based on the measured viability. |
