Provided are a mutant protein of glycosyltransferase and uses thereof. Specifically, by using homology alignment and site-directed mutagenesis, a new mutant protein of glycosyltransferase is discovered. The mutant protein is a non-natural protein with glycosyltransferase catalytic activity, which contains the following core amino acids related to enzyme catalytic activity: the amino acid at position 142 is A, the amino acid at position 144 is H or F and the amino acid at position 339is G, wherein the number of the amino acid position is based on the sequence shown in SEQ ID NO: 13.
