PROBLEM TO BE SOLVED: To provide compositions and methods for targeted cleavage of cellular chromatin in a region of interest, and/or homologous recombination at a predetermined region of interest, in plant cells.SOLUTION: A method for intramolecular homologous recombination in the genome of a plant cell comprises bringing a DNA segment in the genome of the plant cell into contact with a zinc finger nuclease, wherein the DNA segment comprises a first sequence, a second sequence homologous to the first sequence, and a third sequence present between the first sequence and the second sequence; the nuclease cleaves the DNA segment in the third sequence; and the DNA between the first sequence and the second sequence is removed from a chromosome.SELECTED DRAWING: None |
