The present invention relates to an insulinotropic peptide derivative with a modified N-terminal charge and a pharmaceutical composition comprising the same and, more specifically, to an insulinotropic peptide derivative having the N-terminal positive charge modified from neutral pH into a neutral charge or a net negative charge. The insulinotropic peptide derivative according to the present invention shows increased insulin secretion ability and blood glucose lowering ability compared to a natural type insulinotropic peptide while maintaining the stability in blood by rapidly proceeding the dissociation from a GLP-1 receptor due to the modification of the N-terminal charge, thus can be very useful in the treatment of type 2 diabetes.
