<p>A primer set of NKG2D ligands for multiplex RT-PCR(reverse transcription-polymerase chain reaction) is provided to predict sensitivity of a patient to NK(natural killer) cell cancer immunotherapy by analyzing expression of NKG2D ligands in the cancer tissue, and detect amplification error of multiplex RT-PCR. The primer set of NKG2D ligands for multiplex RT-PCR comprises at least two primer pairs selected from a primer pair of SEQ ID NOs:5 and 6 for amplifying MICA(MHC class I-related chain A), a primer pair of SEQ ID NOs:7 and 8 for amplifying MICB(MHC class I-related chain B), a primer pair of SEQ ID NOs:9 and 10 for amplifying ULBP1(UL-16 binding protein 1), a primer pair of SEQ ID NOs:11 and 12 for amplifying ULBP2(UL-16 binding protein 2) and a primer pair of SEQ ID NOs:13 and 14 for amplifying ULBP3(UL-16 binding protein 3). The primer set for detecting amplification error of multiplex RT-PCR comprises a primer pair of SEQ ID NOs:15 and 16 for amplifying a short amplicon of beta-action as a loading control and a primer pair of SEQ ID NOs:3 and 4 for amplifying the whole amplicon of ribosomal protein L19 as a RNA-degradation marker.</p>
