The present invention provides a method of determining the presence and/or amount of osteopontin with intact thrombin-sensitive site in a sample which makes use of an inhibition type assay and a binding agent specific for the thrombin-sensitive site of 5 osteopontin. The present invention also refers to a method of diagnosing or prognosing a disease in a subject, in particular a human, which utilizes an inhibition type assay and a binding agent specific for the thrombin-sensitive site of osteopontin. The present invention further provides a kit comprising an isolated antibody and immobilized osteopontin and the use of the kit for diagnosis or prognosis. 10 The combination of the inhibition type assay along with a binding agent specific for the unique thrombin-sensitive site of osteopontin provides a novel, advantageously fast, advantageously accurate and highly reproducible way for determining the presence and/or amount of osteopontin in particular for diagnosis or prognosis of one of cancer, autoimmune diseases or infections.
