PROBLEM TO BE SOLVED: To provide methods and compositions for inducing pluripotency in differentiated mammalian cells.SOLUTION: The present invention relates to a method for producing a pluripotent stem cell (iPS cell) from differentiated cells, the method comprising: (a) culturing the differentiated cells in a vessel; (b) mechanically agitating the differentiated cells to form a cellular aggregate; (c) exposing the cellular aggregate to an effective amount of a compound having any one of compositions of compounds derived from programin which is a derivative of pyrimidine or purine; (d) detecting the expression level of Oct4, Sox2 or Nanog in the cells of the aggregate; (e) determining that any cell is a pluripotent stem cell if the expression level of Oct4, Sox2 or Nanog in the cell of the aggregate is higher than the corresponding expression level in a differentiated cell; and (f) isolating the pluripotent stem cell.SELECTED DRAWING: Figure 1 |
