摘要 PROBLEM TO BE SOLVED: To provide a cholesterol esterase exhibiting high decomposition activity against a variety of fatty acid sterol esters and having an efficiently synthesizing ability for various sterol esters, and a production method thereof. SOLUTION: Provided is a sterol ester synthetase (cholesterol esterase) having the following characters: (1) mode of action: the synthetase directly acts on sterol and free fatty acids or derivatives thereof without needing coenzymes to catalyze the formation of sterol ester; (2) substrate specificity: the synthetase acts on free fatty acids and sterol or derivatives thereof as a substrate; (3) molecular weight: molecular weight determined by gel filtration method and SDS-PAGE method is 53 kDa to 58 kDa; (4) effective pH and optimum pH: effective pH is 2.5 to 9.5 and optimum pH is 6 to 7.5; and (5) pH stability: the synthetase maintains 75% or more enzymatic activity at pH3.5 to 7.5 in the case where the synthetase is dissolved in various buffer solution of various pH values to allow to stand for 18 hours at 30°C.
