PROBLEM TO BE SOLVED: To provide a method for producing (S)-N-protected propargylglycine represented by formula (2) and having high optical purity. SOLUTION: The production method comprises asymmetrically hydrolyzing R<SP>1</SP>OCO- of an N-protected propargylglycine ester represented by formula (1) [R<SP>1</SP>is ethyl or the like; R<SP>2</SP>and R<SP>3</SP>are each hydrogen or a protecting group of amino (both of R<SP>2</SP>and R<SP>3</SP>are not simultaneously hydrogen); an asterisked carbon atom represents an asymmetric carbon atom] with an enzyme (e.g. derived from the genus Humicola or the genus Pseudomonas).
